Many pharmaceutical compounds are ionic in nature and difficult to retain on conventional ODS columns. A common solution for this is to use ion-pair chromatography (ex: perfluorocarboxylic acids for cations and tetraalkylammonium salts for anions ). Though ion-pair chromatography can be useful, it suffers from the following limitations: incompatible with quantitative LC-MS analysis (ion suppression), long re-equilibration time, and fails to provide sufficient retention for both anions and cations.
These limitations can be overcome with Scherzo SM-C18 (ODS + anion ligands + cation ligands). By utilizing the multi-mode separation (RP + IEX or NP + RP), researchers can now retain and separate ionic polar solutes without ion-pairing additives. In addition, column selectivity can be altered by changing eluent pH. This provides an additional tool for separation scientists. Below are applications for pharmaceuticals:
